Zeiss LSM700

Modalities: invert, confocal, live cell

Overview

The LSM700 is a point scanning confocal microscope built on an inverted microscope body.

There are a full range of objectives and the microscope is automated except for the condenser arm and stage. The focus drive can be driven by the software, making z stacks possible.

For confocal imaging, there are three excitation laser lines (405, 488 and 561 nm) and for detection, two photomultiplier tubes (PMTs) in the scanhead using a shared pinhole which can be separated by a sliding dichroic, plus a transmission PMT for brightfield.

Two or three colour fluorescence imaging is therefore easy, and four or five colour imaging too, provided that good separation of the fluorophores is possible.

Finally, for short term live cell imaging, a stage top incubator is available, with temperature and CO2 regulation.

Imaging modes:

  • Spot scanning, line scanning, 2D (xy) or 3D (xyz)
  • multichannel with timelapse,
  • ROI scanning with bleaching, allowing FRAP
