I joined the Fibrosis research group in September 2014 to complete my PhD with Professor Fiona Oakley. During my PhD I aimed to uncover the cell specific role of the NF-kB subunit c-Rel in tissue repair, fibrosis and cancer utilising several cell specific knockout mice as well as a novel preclinical human precision cut tissue slice system. I identified c-Rel as a critical regulator of metabolic reprogramming during epithelial dedifferentiation and macrophage polarization and that targeting c-Rel therapeutically in murine and human models of fibrosis in multiple organs was protective.
Following completion of my PhD in Sept 2017, I was awarded a Newcastle University Academic Development Scholarship which allowed me to continue my work in the Fibrosis Research Group and finalise my work ready for publication.
In January 2018, I commenced a joint Research Associate position between the Beatson Institute for Cancer Research in Glasgow and Newcastle University aiming to define the role of neutrophils in liver cancer.
Neutrophils are the most abundant immune cell accounting for roughly 60% of all white blood cells in the circulation. It was typically thought that neutrophils were short lived during the inflammatory response, functioning only as pathogen killers. However, it is now appreciated that neutrophils are much longer lived than first thought and are plastic cells that exist on a spectrum of activation and are critical for shaping the immune response. Much of our current understanding of neutrophil plasticity has been identified in models of cancer. Two neutrophil subsets have been identified in cancer; pro- and anti- inflammatory neutrophils termed N1 and N2 respectively.
Our current CRUK funded research aims to investigate the mechanistic basis for the pro-tumour role of neutrophils in liver cancer with the aim to find novel therapies that modulate neutrophil phenotype and the tumour microenvironment to increase the effectiveness of current immunotherapies.
We employ complex genetically engineered mice and multiple dietary and inducible models of liver cancer and multi-omics approaches to dissect the contribution of neutrophils to liver cancer development. As a result of our investigations and the preclinical data generated, we have identified a novel small molecule inhibitor of neutrophil infiltration that in combination with a approved immunotherapy improves therapy response. As a direct result of this we have recently been awarded a CRUK clinical trial award to perform a phase I/IIb clinical trial in liver cancer.
I am currently a named co-supervisor for one PhD student. I am regularly involved in supervising students including nine undergraduate and two master’s students to completion as well as two clinical academic fellows.
